The drive in numbers
From Cairns to Cape Tribulation is about 140 to 145 km, or roughly two and a half to three hours of driving without stops. The first stretch follows the coast road past Palm Cove to Port Douglas, then heads inland past the cane fields at Mossman to the Daintree River. Cape Tribulation is about 35 km beyond the river crossing.
The road is sealed the whole way, so a 2WD hire car is fine. North of the river it narrows and winds through the forest. The Cape Tribulation Road is open to all vehicles as far as just past Cape Trib Beach House. Beyond that the Bloomfield Track is 4WD only, unsuitable for trailers and caravans, and often closed in the wet season. The CREB Track is currently closed.
Add those numbers up and a return trip to Cape Tribulation is five to six hours at the wheel before you have stopped anywhere. A tour covers the same distance; you simply are not the one driving.

Private cars cannot drive up to the gorge. You park at the Mossman Gorge Cultural Centre and take the shuttle bus, which runs every 15 minutes from 8am to 5pm, with the last bus back at 4:30pm. There is a shuttle fare per person. The centre advises against swimming in the Mossman River because of strong currents and submerged objects; if you do go in, it is only in the sheltered spot in the dry season, where staff direct you.
The river cruise
This is the piece to book ahead. The Daintree River Wildlife Experience Cruise costs $25, runs about an hour and includes morning or afternoon tea, rated 4.7 from 521 reviews. The listing recommends hiring a car, since there are no transfers. The Crocodile Express cruise at $28 runs two separate routes on the southern side of the river, and its ticket is a 14-day pass, so you can take the second route another day without crossing on the ferry. The ferry
The Daintree River ferry is a cable ferry that runs every day from 5am to midnight. You pay a per-vehicle fare, set by Douglas Shire Council. River and tidal conditions can occasionally affect it, so allow some slack in the afternoon.
North of the river
Alexandra Lookout, the boardwalks, the Daintree Ice Cream Company and the beach at Cape Tribulation are all on or just off the main road. Stingers are in the water roughly November to May, and crocodiles live along this coast all year, so the beach is for walking.
After dark
The Cape Tribulation Rainforest Night Walk ($39, rated 4.7) is a guided torchlight boardwalk walk, and it only works for self-drivers because no transfers are offered. As a day trip it means driving back to Cairns in the dark after the ferry, so most people who book it stay a night north of the river.

The car and the ferry are shared, so the more people in the car, the further ahead you come out. For one traveller the gap closes; for a family of four it widens. The part nobody plans for: the drive home
The drive up is the easy half. You leave fresh, the coast road is lovely in the morning light, and there is plenty to look at. The drive back comes after a day of walking in heat and humidity, on a narrow, winding road through the forest, then the long run back down the coast to Cairns. In the wet season, add rain.
Tour reviewers mention motion sickness on the bends even as passengers. If your group has one confident driver and no one to share the wheel, a tour is the safer choice for a same-day return.
